In a flash of light: Combining native mass spectrometry with X-ray free electron lasers

One of the main scientific drivers and promises of X-ray free electron lasers (XFELs) has been the possibility to perform single particle diffractive imaging (SPI) without the need for crystallization and thus study transient and low-abundance species. However, currently the biggest obstacle limiting the usefulness of XFEL SPI for structural determination has been the sample introduction into the FEL beam. This contribution will present the work of the interdisciplinary MS SPIDOC consortium, which works toward a novel sample introduction system based on native mass spectrometry for use at beamlines of the European XFEL and other advanced light sources.

Using hydrogen/deuterium exchange to study protein interactions and dynamics

Hydrogen/deuterium exchange mass spectrometry allows the monitoring of protein interactions and structural changes under virtually any conditions. It lacks residue resolution, but this is compensated for by its speed and ability to work in complex environments and with large protein complexes. This talk will give an overview of selected HDX-MS projects. We will look at the analysis of complex, heterogeneous haptoglobin:haemoglobin interactions and show how methodological advances are helping to analyse these proteins even in the context of whole human serum. The second case will demonstrate the structural analysis of cis-prenyl transferase and in the third part we will talk about the structural changes underlying H+/Cl− transport and prokaryotic antiporter.
