Structures in Solution: BioSAXS at BM29 ESRF

BM29 is a highly automated SAXS beamline with two sample environments predominately designed to investigate the structures of biological molecules in solution: sample delivery robot and online SEC. In addition, the newly developed versatile sample exposure unit allows investigation of gels or viscous fluids and also the use of flow or mixing 3D printed cells, a WAXS extension is under preparation. Some examples of research carried on will be shown.
Cryo-EM data processing in CCP-EM Doppio

The CCP-EM suite provides widely used software for cryo‑electron microscopy. The scientific programs are available within a modern web-based graphical user interface: CCP‑EM Doppio. It provides three major workflows: single particle reconstruction, sub-tomogram averaging and atomic model building. These workflows use a range of internal and external programs including RELION for image processing and tools from CCP4 for atomic model fitting, refinement and validation. In this talk, I will provide a brief demonstration of the software’s capabilities. I will also discuss how it was used in a recent study of the structure and function of a bacteriophage integrase complex, highlighting how CCP-EM Doppio eases the processing of challenging data sets.
